In your previous devices, did you use Mi Flash Tool to flash ROMs? If you did, then you should take care not to select Flash and Lock, which is the default by the way. Mi Flash tool flashes ROMs from fastboot. Flashing custom ROMs is from custom recovery. Sent from my MI 9 (Cepheus) using Tapatalk

The answer to your question is, no, flashing firmware in TWRP won't lock your bootloader. Never lock your bootloader if you are unsure, and double check your settings before clicking on that "Start" button in Mi Flash Tool.
