What can I do?

EDIT: I finally made the MiFlash to work ... it seemed that the Qualcom driver wasn't installed properly ...

So now what? Do I need to apply for bootloader unlocking again?

Yes you need to apply.I think that you only can apply once every a month .
After that install TWRP_Leo_7.0.zip via adb.
